Research

The linear genome of higher organisms are known to be folded into chromosomal territories within the three-dimensional (3D) nuclear. Recent achievements on 3D genome organization have revealed frequent chromatin loops within topological-associated domains (TADs) and have implicated general mechanism of chromatin loops between gene promoters and enhancers for transcriptional regulation. Rice (Oryza sativa) is one of the most important food crops in the world and has been established as a model plant for biological research. To explore 3D genome structures and its dynamic (4D) in rice, we have been focused on developing plants 3D genome mapping technologies and exploring mechanism of 3D genome structure on transcription regulation since 2013. More specifically, we applied cutting-edge 3D genome mapping technologies and characterized genome-wide DNA-DNA, RNA-DNA interactions mediated by active gene promoters and heterochromatin at various developmental stages and examined the comprehensive hierarchical 3D genome structure and their dynamics as well as their impacts on transcriptional regulation.
